- 以叶片为外植体建立多花黄精组织培养快速繁殖技术体系,通过添加不同浓度6-BA、NAA、2,4-D和蔗糖的MS培养基中培养,筛选愈伤组织诱导、愈伤组织增殖、丛生芽诱导、丛生芽生根诱导的最佳培养基。结果表明:愈伤组织诱导的最佳培养基为MS+6-BA 3.0 mg·L-1+NAA 4.0 mg·L-1+蔗糖20 g·L-1,愈伤组织诱导率最高为53.89%,生长情况最好;愈伤组织增殖培养的最佳培养基为MS+6-BA 2.0 mg·L-1+NAA 3.0 mg·L-1+蔗糖20 g·L-1,增殖倍数7.87,生长情况相对最好;丛生芽诱导培养的最佳培养基为MS+6-BA 2.0 mg·L-1+NAA 4.0 mg·L-1,出芽率最高为86.11%,生长情况最好,芽高最大为1.70 cm;丛生芽生根诱导的最佳培养基为MS+NAA 1.0 mg·L-1,生根率最高为66.67%,根数、根长和根粗最好,分别为31.0条、0.46 cm和1.100 mm。以叶片为外殖体可短时间获得大量多花黄精组培苗,解决种植生产上的种苗短缺问题。
- Abstract:
- To construct a rapid propagation system of the tissue culture of Polygonatum cyrtonema Huad.which regeneration from the leaves.The aim was to screen the optimum combination for the best medium of callus induction,callus proliferation,cluster bud induction and rooting induction of clustered shoots by adding different 6-BA,NAA,2,4-D and sucrose proportions in MS medium.The results showed that the best medium for callus induction was MS+6-BA 3.0 mg·L-1+NAA 4.0 mg·L-1+sucrose 20 g·L-1,and the callus induction rate was the highest,it reached 53.89%,and the growth was the best.The best medium for callus proliferation was MS+6-BA 2.0 mg·L-1+NAA 3.0 mg·L-1+sucrose 20 g·L-1,and the multiplication multiple was 7.87,and the growth was the best.The best medium for the induction culture of cluster buds was MS+6-BA 2.0 mg·L-1+NAA 4.0 mg·L-1,and the germination rate was the highest,it reached 86.11%,and the maximum height of 1.70 cm.The best medium of rooting induction for clustered shoots was MS+NAA 1.0 mg·L-1,and the rooting rate was the best,it reached 66.67%,and the best root number,length and diameter were 31.0,0.46 cm and 1.100 mm,respectively.A large amount of Polygonatum cyrtonema Huad.tissue culture seedlings could be obtained by the regeneration from the leaves,in a short time,which could solve the shortage of seedlings in planting production.
