- 以紫花补血草叶片为试验材料,采用不同的外植体消毒方法、不同激素配比的初代、继代和增殖培养基以及生根培养基,研究紫花补血草叶片的植株再生和快速繁殖方法。结果表明:紫花补血草的叶片外植体消毒以75%的酒精消毒10 s加0.1%氯化汞浸泡8 min为宜;叶组织脱分化成芽(初代培养)的适宜培养基为MS+BA 1.0 mg/L+NAA 1.0 mg/L;而芽的增殖和生根分别以MS+BA 0.1 mg/L+NAA 0.1~0.5 mg/L和1/2MS+NAA 0.1 mg/L+蔗糖20 g/L较优。通过叶片植株再生,建立了紫花补血草的组培快繁体系。
- Abstract:
- The leaves of Limonium franchetii were used as the explants and cultured in MS media with different portions of hormone to induce plant regeneration.The results showed that the explants sterilizing with 75% alcohol for 10 seconds and 0.1% HgCl2 for 8 minutes yielded good results.The optimal medium for leaf dedifferentiation and clump bud induction was MS+BA 1.0 mg/L+NAA 1.0 mg/L,while the preferable medium for the plantlets rapid propagation was MS+BA 0.1 mg/L+NAA 0.1-0.5 mg/L,and the most favourable medium for adventitious root induction was 1/2MS+NAA 0.1 mg/L+sucrose 20 g/L.Based on the above optimized mediums,a micropropagation system of Limonium franchetii was successfully developed.
