小型西瓜花药愈伤组织诱导及防褐化研究

- 以8份不同基因型的小型西瓜为试材,研究了基因型、植物生长调节剂对花药愈伤组织诱导的影响及暗培养和抗坏血酸对花药褐化的影响,以期为西瓜单倍体育种提供参考依据。结果表明:8份西瓜材料中7个能够形成愈伤组织,愈伤组织诱导率为3.9%~35.6%;不同基因型对激素的响应不同,‘P15’在D5号培养基中(MS+KT 1.0 mg?L-1+NAA 0.5 mg?L-1)愈伤组织诱导率最高,‘M30’和‘M34’在D6号培养基中(MS+KT 1.0 mg?L-1+NAA 1.0 mg?L-1)出愈率最高;黑暗培养48 h对降低西瓜花药褐化作用最好;在诱导培养基中添加1.0 g?L-1 抗坏血酸可降低花药褐化率。
- Abstract:
- The anthers of eight genotypes watermelon were used as test materials to investigate the effect of genotype and plant growth regulator on callus induction,and effect of dark culture and ascorbic acid on anther browning,in order to provide reference for the haploid breeding of watermelon.The results showed that 7 of 8 watermelon materials could form callus and the callus induction rate was from 3.9% to 35.6%.The response of different genotypes to hormones was different,and the callus induction of ‘P15’ was the highest in the D5 medium (MS+KT 1.0 mg?L-1+NAA 0.5 mg?L-1),and the rate of ‘M30’ and ‘M34’ was the highest in the D6 medium (MS+KT 1.0 mg?L-1+NAA 1.0 mg?L-1).The dark culture for 48 hours was the best to reduce the browning effect of watermelon anther.The addition of 1.0 g?L-1 ascorbic acid in the induction medium could reduce the rate of anther browning.
