秋水仙素对平菇单核菌株生长的影响

- 以平菇菌株“灰美2号”的单核菌株为试验材料,利用5个浓度梯度的秋水仙素溶液对单核菌株的菌丝体进行诱变处理,通过拮抗试验、菌丝生长情况、出菇试验、出菇后孢子核酸含量的测定和出菇菌株菌丝形态的观察,探究不同浓度秋水仙素对平菇菌株的影响,以期为秋水仙素食用菌诱变育种提供参考依据。结果表明:不同浓度秋水仙素处理的菌株与CK菌株存在不同程度的拮抗现象;0.01%秋水仙素处理菌株的菌丝生长速率最快,且菌株菌丝浓密,边缘整齐;出菇试验表明各处理后菌株的产量均明显低于CK,其它农艺性状以及核酸含量与CK无明显差异,同时发现平菇单核菌株出菇后菌丝变为双核菌丝,但由单核菌株出菇后变为双核菌丝的菌株农艺性状明显低于双核菌株。分析发现,0.01%~2.00%浓度秋水仙素处理的平菇单核菌株的农艺性状和核酸含量并未发生变化,但较低浓度的秋水仙素可以加快菌丝生长速度。
- Abstract:
- Pleurotus ostreatus monokaryons strains ‘Huimei No.2’ was mutagenized with five concentration gradients of colchicine.To explore the effect of different colchicine concentrations on Pleurotus ostreatus monokaryons strains,the treated strains were investigatedby means of antagonistic experiment,growth of mycelium,fruiting test,and spores nucleic acid content after fruiting,and observation on mycelial morphology of mushroom strains,in order to provide reference for mutation breeding of colchicine edible fungi.The results showed that there was inordinately antagonism between the mutagenesis strain with the CK,and the strains (treated by 0.01% colchicine) had the fastest growth rate,and the mycelia were dense with neat edges.The fruiting test showed that the yield of the treated strains were significantly lower than CK,while there was no significant difference in other agronomic traits and spores nucleic acid content from CK,and Pleurotus ostreatus
