- 以患根腐病的草莓植株为试材,采用组织分离法获得了一株菌株,通过分子生物学技术对该菌株进行鉴定,并测定了其菌丝生长的最佳条件;同时,通过平板对峙法研究了解淀粉芽孢杆菌Y-48、Lh-1和CM3对此病原菌的抑制作用,以期为根腐病病原菌分离及生防菌株筛选等提供参考依据。结果表明:通过18S rDNA序列分析鉴定,菌株BL2016-6为落叶松葡萄座腔菌;菌株BL2016-6生长的适合温度为20~30 ℃,25 ℃为最适温度,菌丝由白色变为灰色,最终变成黑色,菌株致死条件为为70 ℃,处理20 min;解淀粉芽孢杆菌Y-48、Lh-1和CM3均对此病原菌有抑制作用,抑菌率分别为64.6%、62.2%和57.9%。
- Abstract:
- The root of strawberry infected by root rot was used as material,the strain was obtained by using the method of tissue isolation,and was identified based on the molecular biological technology,and the optimum growth conditions of mycelium were determined.At the same time,the inhibitory effect of Bacillus amyloliquefaciens Y-48,Lh-1 and CM3 on the strain was studied by plate confrontation method,in order to supply references for root rot bacteria isolation and screening antagonistic bacteria.The results showed that the strain named that was identified as Botryosphaeria laricina by 18S rDNA sequence analysis.The temperature range for strain BL2016-6 growth was 20-30 ℃,with the optimum temperature at 25 ℃.The color of mycelial changed from white to gray,eventually turned black.In addition,the lethal temperature for mycelial growth was 70 ℃ for 20 minutes.The results of antibacterial test showed that the inhibitory effects of Bacillus amyloliquefaciens Y-48,Lh-1 and CM3 on the strain BL2016-6 were 64.6%,62.2% and 57.9%,respectively.
