番茄子叶和下胚轴离体再生体系的建立

- 摘 要:以‘09-22’番茄无菌苗子叶和下胚轴为外植体,研究了其在MS附加不同浓度6-BA 和NAA/IAA的培养基上的诱导分化及不定芽生根情况。结果表明:IAA比NAA更利于该品种子叶的再生;在不同激素组合下,番茄子叶和下胚轴愈伤组织诱导率均达100%,但子叶的愈伤组织生长状态好于下胚轴,不定芽分化率也略高于下胚轴,然而,下胚轴来源芽的生根较快,且生根效果好于子叶来源芽。该研究筛选出‘09-22’番茄子叶和下胚轴不定芽再生的最佳培养基配方为MS+6-BA 1.0 mg/L+IAA 0.5 mg/L,最佳生根培养基配方为MS+IAA 0.1 mg/L。

- Abstract:The cotyledon and hypocotyl from tomato seedlings ‘09-22’were used to regenerate on the media supplemented with different 6-BA and NAA/IAA concentrations.The results showed that IAA was more favorable for bud regeneration than NAA.Callus induction rate reached up to 100% for both explants,but callus from cotyledon grew better than that from hypocotyl.Furthermore,the rate of adventitious bud differentiation of cotyledon was slightly higher than that of hypocotyl.However,shoots from hypocotyl rooted quicker and better than that from cotyledon.The optimum medium for adventitious bud differentiation was MS+6-BA 1.0 mg/L+IAA 0.5 mg/L.And MS+IAA 0.1 mg/L was the best rooting medium.
