- 以5个不同品种柑桔为试材,研究比较了不同培养基激素浓度、接种外植体类型及不同品种实生试管苗下胚轴对柑桔离体培养外植体愈伤及胚性愈伤诱导分化的影响。结果表明:下胚轴外植体胚性愈伤诱导效果以BA1.0 mg/L+NAA 0.5 mg/L为好,而子叶外植体则以BA 2.0 mg/L+NAA 1.0 mg/L或更高激素浓度条件为宜;外植体胚性愈伤诱导能力以下胚轴最强,子叶与幼根外植体次之,上胚轴最弱。5个供试柑桔品种下胚轴胚性愈伤诱导能力“本地早蜜桔”最强,其次是“四季桔”、“南丰蜜桔”,再次为“刘本橙”,“罗浮金弹”最弱。
- Abstract:
- Taking 5 different varieties of citrus as materials,the effect of different hormone concentrations,explants and varieties on embryogenic callus induction of citrus in vitro were studied.The results showed that the optimal hormone concentration media for inducing embryogenic callus from hypocotyls was MT+BA 1.0 mg/L+NAA 0.5 mg/L,while that for cotyledons was MT+BA 2.0 mg/L+NAA 1.0 mg/L or much higher;the ability of forming embryogenic callus from explants were hypocotyls>cotyledons,rootlets>epicotyls;and the ability of induced embryogenic callus from hypocotyls for citrus varieties were ‘Bendizao mandarin’>‘Calamondin’,‘Nanfeng mandarin’>‘Luiben orange’>‘Oval kumquat’.
