- 以板栗花芽为试材,研究了建立其花芽中吲哚乙酸(IAA)、脱落酸(ABA)、玉米素核苷(ZR)、赤霉素(GA3)4种内源激素含量的定性、定量分析方法。结果表明:采用80%的预冷甲醇水溶液浸提板栗枝芽中内源激素IAA、ABA、GA3、ZR,提取效果较好;采用SPE-C18小柱固相萃取技术进行纯化,采用Symmetry 300TM C18(5 μm,4.6 mm×250 mm),以甲醇:0.075%冰醋酸(90∶5,V∶V)为流动相,在流速1.0 mL/min、柱温30℃,检测波长219 nm的条件下,进行梯度洗脱,分离效果理想,加标回收率达到89.92%~105.30%;该方法准确、灵敏、重现性好。
- Abstract:
- With Chinese chestnut floral bud as materials,an analytical method based on solid phase extraction-high performance liquid chromatography (SPE-HPLC) was established for the determination of endogenous hormones IAA,ABA,GA3 and ZR in chestnut bud.The results showed that,80% precooled methanol aqueous solution was used to extract the buds endogenous hormones IAA,ABA,GA3 and ZR from chestnut,and the result of extraction was better.Separation was performed on a Symmetry 300TM C18 column (5 μm,4.6 mm×250 mm),using a mixture of methano and 0.075% glacial acetic acid solution (90∶5,volume ratio) as mobile phase at a flow rate of 1.0 mL/min.The ultraviolet detection was set at 219 nm,column temperature at 30°C,gradient elution,the separating effect was good,and the recovery ranged from 89.92% to 105.30%.The method was proved to be accurate,sensitive and reproducible.
