- 以矮牵牛幼苗茎段为外植体,研究了不同培养基对矮牵牛丛生芽诱导及生根的影响,以及不同基质的生根效果。结果表明:矮牵牛最佳丛生芽诱导培养基为MS+6-BA 2.0 mg/L+NAA 0.1 mg/L+蔗糖30%,pH 5.8,分化率为92.17%;生根培养基为1/2 MS+IBA 0.1 mg/L,生根率达到93.23%。在各种基质中,珍珠岩+蛭石+草炭土组合生根效果最好,并且与琼脂培养基比较更适于移栽。
- Abstract:
- Taking the stem-segment with axillary buds as explants,the effects of mediums with different concentration of plant growth regulator on inducing cluster buds,multiplication coefficient and rooting rate were studied,the effect of different medium without agar on rooting in vitro were also studied.The results showed that the optimal medium for inducing cluster buds was MS+6-BA 2.0 mg/L+NAA 0.1 mg/L+sucrose 30 g/L,pH 5.8,and the ratio of induction was 92.17%,The rooting culture medium was 1/2MS+IBA 0.1 mg/L and the ratio of rooting was 93.23%.In all kinds of mixture media without agar pearlite-vermiculite-peat was better,and it was more suitable for transplanting compare to agar media.
