菊花“千代姬”组培快繁条件优化

- 以菊花“千代姬”的叶片为外植体,对其愈伤组织的诱导、丛生芽的增殖和生根的诱导等不同过程的各种培养基和培养条件进行了研究。结果表明:愈伤组织诱导最适培养基为SH+0.5 mg/L 6-BA+1.0 mg/L NAA,其诱导率为100%。丛生芽增殖培养基为MS+2.0 mg/L 6-BA+0.2 mg/L NAA,诱导生根的培养基为1/2MS+0.1 mg/L NAA,3类培养基均加蔗糖30 g/L,琼脂6.0 g/L,在pH 5.8、光照强度2 500 lx、温度25℃、光照时间12 h/d的条件下培养最适宜。
- Abstract:
- Taking leaf of Chrysanthemum morifolium ‘Qiandaiji’ as material,tissue culture conditions and media for callus induction,cluster buds propagation and rooting were studied in this article.The results showed that the proper medium of callus induction was SH+0.5 mg/L 6-BA+1.0 mg/L NAA and resulted in an induction efficiency of 100%.The cluster buds well grow in MS+2.0 mg/L 6-BA+0.2 mg/L NAA.And the appropriate rooting medium was 1/2MS+0.1 mg/L NAA.All three types of media were contained 30 g/L sucrose and 6.0 g/L agar with pH 5.8.The culture conditions were optimized to 2 500 lx light intensity,25℃ temperature and 12 h light/12 h dark circle.
