- 以2种花色、花型良好,长势优异的雄性不育百合‘5-4’与‘5-35’为试材,采用组织培养法,研究不同消毒时间、激素浓度及不同基质配比对2种雄性不育百合组织培养过程中的影响,以期建立快速繁殖体系,为无花粉百合的大量生产提供参考依据。结果表明:最佳外植体灭菌方式为75%乙醇消毒20 s,10% NaClO处理8 min,和75%乙醇消毒30 s,10% NaClO处理6 min;最佳诱导培养基为MS+NAA 1.0 mg?L-1+6-BA 0.5 mg?L-1和MS+NAA 1.5 mg?L-1+6-BA 0.5 mg?L-1;最佳增殖培养基均为MS+6-BA 1.5 mg?L-1+NAA 0.5 mg?L-1;最佳生根培养基为1/2MS+IBA 0.1 mg?L-1+NAA 0.1 mg?L-1+蔗糖40 g?L-1和1/2MS+IBA 0.2 mg?L-1+NAA 0.1 mg?L-1+蔗糖40 g?L-1;最佳移栽基质比例为蛭石∶珍珠岩∶草炭=1∶1∶1。
- Abstract:
- Taking two kinds of male sterile lilies ‘5-4’ and ‘5-35’ with good color,flower type and excellent growth as test materials,tissue culture methods were used to study different disinfection times,hormone concentrations and different substrate formulation on the tissue culture process of two kinds of male sterile lilies,in order to establish their rapid propagation system and provide reference for mass production of non-pollen lilies.The results showed that the best way to sterilize explants was 75% ethanol disinfection for 20 seconds,10% NaClO solution for 8 minutes,and 75% ethanol for 30 seconds,10% NaClO solution for 6 minutes;the best induction medium was MS+NAA 1.0 mg?L-1+6-BA 0.5 mg?L-1 and MS+NAA 1.5 mg?L-1+6-BA 0.5 mg?L-1;the best proliferation medium was MS+6-BA 1.5 mg?L-1+NAA 0.5 mg?L-1;the best rooting medium was 1/2MS+IBA 0.1 mg?L-1+NAA 0.1 mg?L-1+sucrose 40 g?L-1 and 1/2MS+IBA 0.2 mg?L-1+NAA 0.1 mg?L-1+sucrose 40 g?L-1;the best transplanting substrate ratio was vermiculite∶perlite∶peat=1∶1∶1.
