干旱胁迫对远志愈伤组织次生代谢产物含量及抗氧化酶活性的影响

- 以远志愈伤组织为试材,使用不同浓度PEG 6000溶液模拟干旱胁迫,浓度指标分别为5%、10%、15%、20%、30%,以蒸馏水为对照,研究了干旱胁迫对于远志愈伤组织生长量、总酚、总黄酮、远志·酮Ⅲ含量及抗氧化酶活性的影响,以期为揭示远志愈伤组织应对干旱胁迫的机制和远志愈伤组织次生代谢产物的规模化生产提供参考依据。结果表明:在不同程度的干旱胁迫下,远志愈伤组织生长量、总酚、总黄酮、远志·酮Ⅲ含量及抗氧化酶活性差异较大。随着PEG 6000溶液浓度的增加,远志愈伤组织的鲜质量、干质量、总黄酮含量及过氧化物酶(POD)活性基本呈现先增加后减少的趋势;总酚含量、远志·酮Ⅲ含量基本呈现逐渐降低的趋势;超氧化物歧化酶(SOD)活性呈现先降低后升高的趋势;过氧化氢酶(CAT)活性基本呈现逐渐升高的趋势,说明一定程度的干旱胁迫对于远志愈伤组织生长及其次生代谢产物的积累有一定的促进作用。

- Taking Polygala tenuifolia callus as test material,using different concentrations of PEG 6000 (polyethylene glycol) solutions to simulate drought stress,the concentration indicators were 5%,10%,15%,20%,30%,and distilled water was used as a control.The effects of drought stress on the growth,total phenols,total flavonoids,Polygala tenuifolia ketone Ⅲ content and antioxidant enzyme activity of Polygala tenuifolia callus were studied,in order to reveal the mechanism of Polygala tenuifolia callus responding to drought stress and to provide a certain reference for the secondary growth of Polygala tenuifolia callus and the large-scale production of metabolites.The results showed that under different degrees of drought stress,the growth,total phenols,total flavonoids,Polygala tenuifolia ketone Ⅲ content and antioxidant enzyme activity of Polygala tenuifolia callus were quite different.With the increasing of the concentration of PEG 6000 solution,the fresh mass,dry mass,total flavonoid content and POD activity of Polygala tenuifolia callus basically increased first and then decreased;total phenol content,Polygala tenuifolia ketone Ⅲ content basically showed a gradually decreasing trend;SOD activity showed a trend of first decreasing and then increasing;CAT activity basically showing a gradually increasing trend,indicating that a certain degree of drought stress had a certain promoting effect for the growth of callus and the accumulation of its secondary metabolites.
