聚乙二醇提高老化白菜种子活力的研究

- 为探究聚乙二醇处理对老化白菜种子活力提高的作用程度及效果,以“东白一号”白菜种子为试验材料,采用L9(3)4正交实验设计方法对其进行处理,探究了PEG不同分子量、不同处理浓度和不同处理时间的作用条件下对老化白菜种子萌发活力的影响,并进一步测定PEG处理老化种子活力提高后的可溶性蛋白质含量、可溶性糖含量、丙二醛(MDA)含量及抗氧化酶系统活性,以获得不同处理组合对老化白菜种子活力提高的程度并探究种子活力提高背后的机制。结果表明:PEG处理可以明显提高种子活力;以PEG分子量10 000、浓度25%、处理时间8 h下引发的老化白菜种子提高种子活力的效果最好,其发芽率、发芽势及活力指数相比对照(CK)分别高出19.00%、13.67%、571.2,种子活力的影响因素主次顺序为浓度>分子量>处理时间。
- Abstract:
- In order to investigate the effect of PEG on improving seed vigor of aged Chinese cabbage.The orthogonal test method was used to treat the aged Chinese cabbage seeds ‘Dongbai No.1’ as materials to explore the effects of different molecular weight,concentration and treatment time of the PEG on the vigor of aged Chinese cabbage seeds.Then,the contents of soluble protein,soluble sugar,MDA and the activity of antioxidant enzymes were measured,which were the factors of the aged Chinese cabbage seeds after treated with PEG.In order to obtain the degree of improving seed vigor of aged Chinese cabbage by different treatment combinations and explore the mechanism behind the improvement of seed vigor.The results showed that the PEG treatment could significantly improve seed vigor.In this experiment,the best combination of improving seed vigor was that the aged Chinese cabbage seeds induced by PEG with molecular weight of 10 000,concentration of 25% and treatment time of 8 hours.Comparing with the control,the germination potential was higher than 19.00%,the germination rate was higher than 13.67%,vigor index was 571.2 higher.The concentration had most influence,the second was molecular weight and the least was treatment time.
