- 以“嘎拉”苹果单芽系组培苗叶片为外植体诱导愈伤组织,研究了愈伤组织诱导过程中植物生长调节剂种类及浓度配比、光照和黑暗条件以及继代天数对其形成及生长的影响。结果表明:各组合均能比较高效地诱导愈伤组织的形成,以MS+蔗糖30 g?L-1+2,4-D 2.0 mg?L-1 +NAA 0.5 mg?L-1+BA 2.0 mg?L-1诱导效率最佳;愈伤组织的形成需要14~21 d的暗培养,继代周期以15~30 d为宜。
- Abstract:
- Apple as the ‘Gala’ single bud somaclone leaf as explant callus induction,callus induction were studied in the process of plant growth regulators types and concentration ratio,light and dark conditions and transgenerational days on its formation and growth.The results showed that the combination could be more efficiently induce the formation of callus,MS + sugar 30 g?L-1+ 2,4-D 2.0 mg?L-1+ NAA 0.5 mg?L-1+ BA 2.0 mg?L-1 induced efficient;the formation of callus needed 14-21 days of dark raise,transgenerational cycle with 15-30 days was advisable.
