猕猴桃花粉生活力测定方法及花药处理方法研究

- 花粉生活力测定与花药前期处理对人工授粉有着至关重要的意义。现以中华猕猴桃为试材,研究猕猴桃花粉生活力最佳的测定方法以及不同处理对花粉活力的影响。结果表明:TTC染色法测定猕猴桃幼蕾期花粉生活力低,而离体萌发法测定的花粉萌发率高,建议在生产中最好采用染色法与离体萌发法综合测定花粉生活力;花粉萌发率与花药脱离花瓣时间有关,花蕾期花朵采摘后应立即分离雄蕊与花瓣,以提高花粉萌发率;花粉干燥与环境条件有关,如环境已适宜干燥花粉,建议不要采用其它辅助干燥花粉的方法。
- Abstract:
- Method of measuring pollen viability and pretreatment of pollen have very important significance for artificial pollination.Taking Actinidia chinensis as materials,the best way of measuring pollen viability and the effect of different treatments were studied.The results showed that,for pollen of flower bud,the method of TTC,the pollen viability was low,and the method of germination in vitro and the pollen viability was high.So,the best way of measuring pollen viability was methods of staining and germination.The rate of pollen germination was associated with the time,and for flower bud,the best way was separation of stamens and petals immediately for improving the rate of pollen germination;Pollen drying was associated with environmental condition,if the environment was suitable for drying pollen,it didn’t need to use other drying methods.
