- 以具有较多优良性状的F1代西瓜品种“春光”、“拿比特”和“喜都”的花药为试材,研究了影响西瓜花药愈伤组织诱导率的因素。结果表明:西瓜花药在MS+KT 1.5 mg/L+BA 2.0 mg/L+NAA 1.5 mg/L上诱导率最高;固体培养时蔗糖浓度以6%效果较好;4℃下低温预处理72 h可以提高西瓜花药愈伤组织的诱导率;30℃下高温预培养72 h后置于25℃低温下培养也可提西瓜花药愈伤组织的诱导率;黑暗条件下比较容易诱导出质地良好的西瓜花药愈伤组织。
- Abstract:
- Using three Citrullus lanatus cultivars of F1generation ‘Chunguang’,‘Nabite’,‘Xidu’ which including lots of merits as materials,the technology of anther culture were studied.The results showed that more callus were formed on MS+KT 1.5 mg/L+BA 2.0 mg/L+NAA 1.5 mg/L;the best concentration of sucrose for callus induction was 6%;the results indicated that the treating of 4℃,72 hours was the proper condition that make the frequencies of callus induction;the studying of culture conditions and culture means indicated that callus inducement was better when it was cultured at 30℃ for 72 h and then cultured at 25℃.The quality of watermelon callus induced in dark was much better than in light.
