- 以涉县柴胡茎叶为试材,采用单因素试验和四因素三水平的正交实验设计,研究优化了总黄酮乙醇回流浸提工艺,并分析测定提取物及与维生素C复配后的抗氧化活性,以期为涉县柴胡的进一步开发利用提供参考依据。结果表明:1)涉县柴胡茎叶总黄酮最佳提取工艺为料液比1∶20 g?mL-1,提取时间2 h,提取温度70 ℃,乙醇浓度60%;2)最优条件下涉县柴胡茎叶总黄酮对DPPH?、?OH、O?2的半抑制浓度分别为0.223、0.413 mg?mL-1和0.445 mg?mL-1;〖JP+1〗3)与维生素C复配后表现出一定协同作用,试验组内复配液(1∶3)协同作用最强。综上可知,涉县柴胡茎叶中总黄酮具有很好的抗氧化能力。
- Abstract:
- Taking the aerial part of Shexian bupleurum as test materials,the ethanol reflux extraction process of total flavonoids was studied and optimized by single factors test and four factors and three-levels orthogonal test.The antioxidant activity of the extract and compound with vitamin C were studied,in order to provide reference for the further development and utilization of bupleurum in Shexian.The results showed that,1) the best extraction process for total flavonoids from aerial part of bupleurum was solid-liquid ratio was 1∶20 g?mL-1,extraction time was 2 hours,extraction temperature was 70 ℃,and ethanol concentration was 60%.2) The half inhibition rate concentration (IC50) of total flavonoids from aerial part of bupleurum against DPPH?,?OH and O?2 radical were 0.223,0.413 mg?mL-1 and 0.445 mg?mL-1,respectively.3) It showed a certain synergistic effect after compounding with vitamin C,the best synergistic effect was compound solution (1∶3) in all experimental groups.In conclusion,the total flavonoids in the aerial part of Shexian bupleurum had good antioxidant capacity.
