- 以樱桃型紫色番茄“砚紫1号”种子为试材,通过无菌处理获得无菌苗,用无菌苗的幼嫩子叶为外植体,研究了诱导愈伤组织分化及生根情况, 以期建立高效的离体培养再生体系。结果表明:紫色番茄无菌苗子叶在MS+IAA 0.1 mg·L-1+6-BA 0.5 mg·L-1培养基条件下的愈伤组织诱导状态最佳;将愈伤组织进行暗培养处理,可诱导愈伤产生不定芽,其中最佳的培养基条件是MS+NAA 0.01 mg·L-1+6-BA 2.0 mg·L-1;诱导再生的不定芽经过2周光培养接种至MS+NAA 0.05 mg·L-1培养基中,诱导生根良好,可获得完整的再生植株。
- Abstract:
- Cherry purple tomato ‘Yanzi No.1’ seeds were taken as experimental materials,and the sterile seedlings were obtained after sterile treatment;and then the immature cotyledons of sterile seedlings were used as explants for callus induction,differentiation and rooting were studied,and a complete tissue culture regeneration system was established.The results showed that purple tomato cotyledon cultured in MS+IAA 0.1 mg·L-1+6-BA 0.5 mg·L-1 was the best under the condition of callus induction medium;callus dark culture could induce callus to produce adventitious buds,the culture medium MS+NAA 0.01 mg·L-1+6-BA 2.0 mg·L-1 was the best;inducing regeneration of adventitious buds were cultured after 2 weeks of light in MS+NAA 0.05 mg·L-1 medium,rooting was fine,a complete regeneration system could be obtained.
