- 以铁皮石斛为试材,采用叶面喷施水杨酸(SA)的方法,测定了铁皮石斛经SA预处理后在低温胁迫下根系活力和保护酶活性的变化,探讨了水杨酸对低温胁迫下铁皮石斛幼苗根系的影响。结果表明:铁皮石斛幼苗根系活力在低温胁迫第10天时较对照显著降低了23.05 μg·g-1?h-1,超氧化物歧化酶(SOD)、过氧化物酶(POD)、抗坏血酸过氧化物酶(APX)和过氧化氢酶(CAT)的活性在低温胁迫第7天时较对照分别显著提高了28.10 U·g-1,140.65、75.05、48.21 U?g-1·min-1。铁皮石斛幼苗经0.5 mmol·L-1 SA预处理后,在低温胁迫第7天时根系活力提高了18.20 μg·g-1·h-1,SOD、POD、APX、CAT活性分别提高了22.54 U·g-1,59.00、64.57、21.81 U·g-1·min-1。这表明适宜浓度的外源SA能够有效缓解低温胁迫对铁皮石斛的伤害,提高其耐低温的能力。
- Abstract:
- Dendrobium officinale Kimura et Migo seedling was used as material,SA(Salicylic acid) was sprayed to D.officinale leaves,changes of root activity and antioxidant enzyme of D.officinale under low temperature stress were observed.To study the effects of SA pretreatment on D.officinale seedlings root under low temperature stress.The results showed that,at the tenth day of cold stress,the root activity in D.officinale seedlings was significantly decreased by 23.05 μg·g-1·h-1.A and the activities of superoxiede dismutase(SOD),peroxidase(POD),ascorbate peroxidase (APX) and catalse(CAT) were significantly increased by 28.10 U?g-1,140.65 U·g-1·min-1,75.05 U·g-1·min-1,48.21 U·g-1·min-1 at the seventh day of low temperature stress,respectively.Petreatment with 0.5 mmol?L-1 SA,D.officinale seedlings root activity significantly increased by 18.20 μg·g-1·h-1,and the activities of SOD,POD,APX and CAT were significantly increased by 22.54 U·g-1,59.00 U·g-1·min-1,64.57 U?g-1?min-1,21.81 U·g-1·min-1 at the seventh day of low temperature stress,respectively.It had been concluded that SA in appropriate can slow down the hurt of low temperature and enhance cold tolerance of D.officinale.
