- 向培养液中分别添加不同浓度的亚硒酸钠、氯化钙,用液体振荡培养法培养蛹虫草菌丝体,用HPLC法测定虫草素与腺苷含量,用苯酚-硫酸法测定多糖含量,用高碘酸钠比色法测定虫草酸含量。以研究2种微量元素对蛹虫草菌丝体中虫草素、腺苷、多糖和虫草酸含量的影响。结果表明:向培养液中添加适量的硒或钙,可显著提高蛹虫活性物质含量。添加15 mg/L亚硒酸钠,虫草素和虫草酸含量比对照组分别提高了29.0%和34.9%;添加16 mg/L氯化钙,虫草素含量比对照提高24.05%。
- Abstract:
- Adding different levels of Na2SeO3 and CaCl2 to the culture medium,with the method of liquid shake culture,the effect of 2 trace elements on the content of cordycepin and adenosine,polysaccharide and cordyceps acid were studied,with HPLC to test the level of cordycepin and adenosine;with phenol-sulphate acid to test the level of polysaccharide;with sodium periodate colorimetric to test the level of cordyceps acid.The results showed that adding suitable selenium or calcium could obviously raised the content of Cordyceps militaris active substances.Adding 15 mg/L Na2SeO3 to the culture medium and the level of cordycepin and cordyceps acid had raised 29.0% and 34.9% respectively.Adding 16 mg/L CaCl2 and the level of cordycepin had raised 24.05%.
