- 以旱半夏为试材,采用秋水仙素浸泡法,研究了不同的秋水仙素浓度和不同的处理时间对旱半夏多倍体成活率和诱导率的影响,以期为旱半夏多倍体的育种研究提供参考依据。结果表明:秋水仙素浸泡法获得旱半夏多倍体的最佳处理条件为25 ℃,0.06%秋水仙素处理4 d。显微观察发现,处理后的旱半夏多倍体出现不同水平的染色体加倍,气孔增大,气孔密度降低。与对照相比,旱半夏多倍体植株的叶片肥厚,植株粗壮,植株高度、叶柄直径、叶片大小、子代块茎直径和质量均显著增加。
- Abstract:
- Taking Pinellia ternata as experimental material,colchicine immersion method was adopted,the effects of different concentrations of colchicine and different treatment times on the survival and induction rates of polyploids of Pinellia ternata were studied,in order to provide reference for the breeding research of Pinellia ternata polyploids.The results showed that the optimal treatment conditions for obtaining Pinellia ternata polyploids by colchicine immersion method were at 25 ℃,with 0.06% colchicine treatment for 4 days.Microscopic observation revealed that the treated Pinellia ternata polyploids exhibited different levels of chromosome doubling,increased stomatal size,and decreased stomatal density.Compared with the control group,the leaves of the Pinellia ternata polyploid plants were thicker,the plants were sturdier,and the plant height,petiole diameter,leaf size,and the diameter and weight of the progeny tubers were significantly increased.
