- 以大花耧斗菜为试材,采用常规石蜡切片显微观测等方法,研究了0%(CK)、10%、20%、30%、40% PEG浓度干旱胁迫对耧斗菜在0、3、6、9、12 d的须根数、根长、根冠比、维根比等形态指标以及可溶性糖、可溶性蛋白质、丙二醛等生理指标的影响,以期从细胞学和生物化学层面分析耧斗菜抗旱机理。结果表明:干旱胁迫对大花耧斗菜须根数无显著性影响;根长显著增加,40% PEG胁迫12 d,根长从20.94 cm增加至25.60 cm;根冠比在胁迫3 d后显著增加,40% PEG胁迫12 d,根冠比从0.846增加至1.952;维根比随着PEG浓度的增加显著增加,PEG胁迫6 d,维根比从0.337增至0.483,PEG胁迫12 d,PEG浓度对维根比影响不显著,但均高于CK的0.359。干旱胁迫下大花耧斗菜植株丙二醛含量和游离脯氨酸含量总体呈上升趋势,可溶性蛋白质含量变化不显著,可溶性糖含量的变化趋势是整体先下降后上升,而且含量都低于CK。
- Abstract:
- The effects of drought stress on the anatomical structure and physiological indexes of the columbine root were studied with the method of conventional paraffin section.Experiment with columbines as experimental material,using paraffin section and microscopic observation of 0(CK),10%,20%,30%,40% PEG concentration roots under drought stress in 0 day,3 days,6 days,9 days,12 days of root number,root length,root cap ratio,V/R ratio,and the change of physiological and biochemical indexes such as soluble sugar,soluble protein and malondialdehyde,so as to analyze the drought resistance mechanism of columbine from the cytological and biochemical levels.The results showed that drought stress had no significant effect on the number of fibrous root of columbine,root length increased significantly,root length increased from 20.94 cm to 25.60 cm under 40% PEG drought stress at 12 days.The root cap ratio increased significantly after stress at 3 d,R/C from 0.846 to 1.952 under 40% PEG drought stress at 12 days;V/R increased significantly with the increase of PEG concentration,V/R increased to 0.483 from 0.337 under PEG drought stress at 6 days.There were no significant effect on V/R under PEG stress at 12 days and PEG concentration,but all V/R were higher than CK 0.359.Under drought stress,the content of malondialdehyde and the content of free proline in the plants of columbine were on the rise,and the change of soluble protein was not significant.The change trend of soluble sugar content was first descended and then increased,and the content of soluble sugar was lower than CK.
