通用稳定向日葵耐盐细胞系的建立

- 以10个向日葵品种为试材,利用细胞悬浮技术,研究了培养耐盐细胞系的最佳条件。结果表明:切取向日葵7日龄无菌苗子叶与胚轴接种于6-BA 05 mg?L-1+NAA 05 mg?L-1+NaCl 50 mmol?L-1+蔗糖40 g?L-1,pH 55~70的MS培养基中,培养温度为28 ℃,14 d后取其1 g愈伤组织接种于添加 NaCl 175 mmol?L-1 的30 mL液体培养基中,振荡培养,7 d继代1次,即可获得稳定耐盐细胞系。该方案不仅适合油葵也适用食葵。
- Abstract:
- Taking 10 varieties of Helianthus annuus L.as test materials,using the cell suspension technology,the optimum conditions for cultivation of salt-tolerant cell lines were studied.The results showed that cutting out the seven-day sunflower′s cotyledons and hypocotyls and inoculating in 6-BA 05 mg?L-1+NAA 05 mg ?L-1+NaCl 50 mmol?L-1+sucrose 40 g?L-1,pH 55-70 MS medium and culturing in temperature at 28 ℃ for two weeks.Then cutting 1 g the callus and put them in the 30 mL liquid medium which contained NaCl 175 mmol?L-1,shaking and subculturing every seven days,the stable salt-tolerant cell lines were obtained.This scheme was not only applicable for oil sunflower but also for eating sunflower.
