- 以“津研4号”黄瓜为试材,采用盆栽试验的方法,设置LB培养基(100%灌根,CK1),枯草芽孢杆菌(100%灌根,TK1),PD培养基(100%灌根,CK2),哈茨木霉(100%灌根,TK2),混合培养基(50% LB培养基+50% PD培养基灌根,CK3),混合菌剂(50%枯草芽孢杆菌+50%哈茨木霉灌根,TK3) 6个处理,通过分析幼苗植株鲜质量及根冠性状研究根系形态,采用WinRHIZO根系扫描仪研究黄瓜根分布及根构型,应用因子分析和Pearson相关分析获取根系构型几何特征之间的相关关系,通过主成分分析法计算主成分得分和综合评分,并利用聚类分析比较不同菌剂处理对黄瓜根系生长及构型响应的差异,研究了枯草芽孢杆菌、哈茨木霉对植物根系的影响,以期为微生物菌剂的合理配施提供参考依据。结果表明:不同菌剂处理对黄瓜幼苗根系比根长、根组织密度、根细度、分枝密度、分形维数无显著影响(P>005);施加枯草芽孢杆菌条件下黄瓜幼苗的根表面积、根组织密度增幅最大,比根长、比根表面积降幅最大;施加哈茨木霉对黄瓜根系根长、根体积、链接数增幅最大,对分枝密度降幅最大;施加混合菌剂对黄瓜地上部干质量、地下部干质量、根冠比、分支数、交叉数增长最大,对比根长、比根表面积降幅最大。相关性分析表明根系构型的主要影响因子是链接数、根表面积和根体积。不同菌剂处理的黄瓜根系综合得分依次为混合菌剂>枯草芽孢杆菌>哈茨木霉>PD培养基>LB培养基>混合培养基。说明混合菌剂处理下黄瓜幼苗根系生长及构型综合评价较高,PD培养基、LB培养基、枯草芽孢杆菌、哈茨木霉处理下根系评价中等,混合培养基处理下根系评价最低。
- Abstract:
- Cucumis sativus L.‘Jinyan No4’ was used as experimental material,a pot experiment was adopted.LB medium (100% root irrigation,CK1),Bacillus subtilis (100% root irrigation,TK1),PD medium (100% root irrigation,CK2),Trichoderma harzianum (100% root irrigation,TK2),mixed medium (50% LB medium+50% PD medium,CK3) and mixed bacterial agents were set (50% Bacillus subtilis+50% Trichoderma harzianum root irrigation,TK3),the root morphology was studied by analyzing the fresh quality of seedling plants and root cap characters,the root distribution and root configuration of cucumber were studied by WinRhizo root scanner,the correlation between the geometric characteristics of root configuration was obtained by factor analysis and Pearson correlation analysis,the principal component score and comprehensive score were calculated by principal component analysis,and the different bacterial agents were compared by cluster analysis.The effects of Bacillus subtilis and Trichoderma harzianum on plant roots were studied,in order to provide a reference for the rational application of microbial agents.The results showed that different bacterial agents had no significant effects on the specific root length,root tissue density,root fineness,branch density and fractal dimension of cucumber seedlings (P>005).Under the condition of Bacillus subtilis,the root surface area and root tissue density of cucumber seedlings increased the most,and the specific root length and specific root surface area decreased the most;the application of Trichoderma harzianum increased the root length,root volume and link number of cucumber roots the most,and the branch density decreased the most;the application of mixed bacterial agent affected the aboveground dry mass,underground dry mass,root shoot ratio,branch number and crossing number of cucumber seedlings.The correlation analysis showed that the main influencing factors of root configuration were link number,root surface area and root volume.The comprehensive scores of cucumber roots treated with different bacterial agents were as follows,mixed bacterial agent>Bacillussubtilis>Trichoderma harzianum>PD medium>LB medium>mixed medium.The comprehensive evaluation value of length and configuration was high,the root evaluation was medium under the treatment of PD medium,LB medium, Bacillus subtilis and Trichoderma harzianum,and the root evaluation was the lowest under the treatment of mixed medium.
