- 从膜荚黄芪中克隆了CHR基因,GenBank登陆号为KY086286(AmCHR),AmCHR全长cDNA为1 173 bp,开放阅读框为957 bp,编码318个氨基酸,其分子量为35.58 kDa,等电点为 6.26。生物信息学分析表明AmCHR蛋白与豆科植物CHR同源,无信号肽,定位于细胞质。实时荧光定量PCR和高效液相色谱分析结果显示AmCHR在根、茎、叶中的表达水平与毛蕊异黄酮葡萄糖苷的含量变化一致。该研究从膜荚黄芪中克隆了1个新的AmCHR基因,推测AmCHR的表达与膜荚黄芪毛蕊异黄酮葡萄糖苷的积累密切相关。
- Abstract:
- A CHR gene was cloned from Astragalus membranaceus in the present study,the GenBank accession number was KY086286 (AmCHR),the full-length cDNA of AmCHR was 1 173 bp and it consisted of 957 bp open reading frame encoding 318 amino acids,which predicated molecular weight was 35.58 KDa and isoelectric point was 6.26.Bioinformatics analysis showed that AmCHR was homology with other plant CHR.Real-time PCR and HPLC indicated that the expression levels of AmCHR was similar to the changes of CG contents in different tissues.These results showed that the cloned AmCHR was a new CHR gene and the expression of AmCHR might be relevant to CG accumulation in A.membranaceus.
