- 以猴头菌0605为试材,采用单因素试验和正交实验,研究了酶系统、酶解时间、酶解温度、菌龄、恒温摇床转速单因素对其原生质体制备与再生的影响。结果表明:原生质体制备和再生的最佳条件为菌龄10 d的菌丝体,在酶系统2.0%溶壁酶+0.5%崩溃酶作用下,32 ℃酶解2.5 h,恒温摇床转速为120 r?min-1。该试验在最佳条件下获得的猴头菌原生质体数量与再生率均较高,为进一步开展猴头菌原生质体融合育种奠定基础。
- Abstract:
- Hericium erinareus 0605 was used as test material,the influence of enzyme system,enzymolysis time,enzymolysis temperature,mycelial age and shaking speed on the preparation and regeneration of protoplasts of Hericium erinareus 0605 were studied by single factors and orthogonal design experiments.The results showed that the optimal preparing and regeneration conditions of Hericium erinareus protoplasts were mycelium age 10 days,using 2.0% lywallzyme and 0.5% driselase,digesting at 32 ℃,shaking speed 120 r?min-1 for 2.5 hours.Under this conditions,the study established the foundation of protoplast fusion breeding of Hericium erinareus.
