- 以抗病杂合体(Ty-2/ty-2)、抗病纯合体(Ty-2/Ty-2)和感病纯合体(ty-2/ty-2)番茄为试材,以提取植物的基因组DNA作为模板,根据与抗病基因Ty-2紧密连锁的RFLP标记设计特异引物,采用DNA聚合酶链式反应的方法,筛选获得了2个与抗病基因Ty-2紧密连锁的、且稳定可靠的SCAR标记,分别命名为T0302-1和T0302-2。结果表明:T0302-1和T0302-2两个标记均为共显性分子标记,可用于番茄抗病育种的辅助选择中。该研究旨在筛选获得与番茄黄化曲叶病毒病抗病基因Ty-2紧密连锁的分子标记,为番茄抗病育种提供技术支撑。
- Abstract:
- Resistant heterozygotes (Ty-2/ty-2),resistant homozygotes (Ty-2/Ty-2) and susceptible homozygotes (ty-2/ty-2) were used as experimental materials.Based on the published RFLP markers tightly linked to Ty-2,three pairs of specific primers were designed for PCR amplification.As a result,two steady-going and effective SCAR markers,T0302-1 and T0302-2,were found.The results showed that T0302-1 and T0302-2 were co-dominant SCAR markers,which could be used in marker assistant selection for tomato disease resistance breeding.This paper was aimed to obtain molecular markers tightly linked to Ty-2 gene resistant to tomato yellow leaf curl virus and afford technical supports for tomato disease resistance breeding.
