苦豆子内生放线菌产酶特性分析及活性菌株鉴定

- 以宁夏干旱荒漠区的126株苦豆子为试材,采用透明圈法对分离自其内生放线菌进行分泌胞外酶活性检测。结果表明:有71株内生放线菌具有酶活性,占总菌株数的56.3%。其中有3株菌株酶活性最为显著,菌株NDZKDS73、NDZKDS7具有的淀粉酶活性最高,透明圈半径达到24.62mm和19.12mm;菌株FJYKDS5具有的蛋白酶活性高,透明圈半径16.94mm;菌株FJYKDS11、BJGKDS2具有的纤维素酶活性最高,透明圈半径为24.72mm和22.40mm;菌株FJJKDS2具有的几丁质酶活性最高,透明圈半径17.76mm;菌株NDZKDS24、NDZKDS20对葡聚糖酶活性最高,透明圈半径为12.06mm和10.50mm。通过形态特征和16SrRNA基因序列同源性分析,有7株活性菌属链霉菌属(Streptomyces),1株属拟诺卡氏菌属(Nocardiopsis)。

- Taking126endophyticactinomycetesfromSophoraalopecuroidesL.inariddesertregionofNingxiaasmaterial,throughtransparentcirclemethod,studiedthefeatureofexcretingenzyme.Theresultsshowedthatthe71strainscouldproduceenzyme,accountingfor56.3%oftotalisolates,inwhich8strainswerethemostsignificantlyactive.Inthesestrains,NDZKDS73 andNDZKDS7strainsproducedthehighestactivityamylase,thetransparentcircleradiuswere24.62mmand19.12mm,respectively;FJYKDS5strainproducedthehighestactivityprotease,thetransparentcircleradiuswas16.94mm;FJYKDS11andBJGKDS2 strainsproducedthehighestactivitycellulase,transparentcircleradiuswere24.72mmand 22.40mm,respectively;FJJKDS2strainsproducedthehighestactivitychitinase,transparentcircleradiuswas17.76mm;NDZKDS24andNDZKDS20 strainsproducedhighestactivityglucanase,transparentcircleradiuswere12.06mmand 10.50mm,respectively.Bymorphologicalcharacteristicsand16SrRNAgenesequencesimilarityanalysis,7strainswereidentifiedas Streptomyces and1strainwere Nocardiopsis in8strains.
