- 以甜瓜品种“黄醉仙”下胚轴为外植体,研究了添加不同浓度的BA和IAA激素组合对其愈伤组织、不定芽的诱导和不定芽伸长的影响,同时研究了添加不同浓度IBA激素组合对其生根的影响。结果表明:甜瓜“黄醉仙”下胚轴诱导愈伤组织最适宜培养基是MS+BA -1.0 mg/L+IAA 0.5 mg/L和MS+BA 1.0 mg/L+IAA 1.0 mg/L,最适宜不定芽诱导的培养基是MS+BA 2.0 mg/L,不定芽伸长的最适宜培养基是MS+IAA 1.0 mg/L,而生根的适宜培养基是1/2MS+IBA 1.0 mg/L。
- Abstract:
- Taking the hypocotyl of melon cv.‘Huangzuixian’ as explants,the effects of MS media with different concentrations of BA and IAA combination on the callus and adventitious bud induction,as well as adventitious buds elongation were investigated.Meanwhile,the effects of MS media with different concentrations of IBA on rooting of regenerative buds were also investigated.The results showed that the optimal medium for inducing melon ‘Huangzuixian’ callus was MS+BA 1.0 mg/L+IAA 0.5 mg/L and MS+BA 1.0 mg/L+IAA 1.0 mg/L,the optimal medium of adventitious bud induction was MS+BA 2.0 mg/L,and the optimal media of adventitious bud elongation was MS+IAA 1.0 mg/L while the optimal rooting media was 1/2MS+IBA 1.0 mg/L.
