- 以10个品种苜蓿无菌苗的子叶和下胚轴为外植体,研究了不同培养基、不同激素种类和配比对愈伤组织诱导率及分化率的影响。结果表明:国产苜蓿下胚轴外植体愈伤组织分化率最高;愈伤组织诱导最适培养基为MS盐+UM维生素+2 mg/L 2,4-D+025 mg/L KT+2 g/L水解酪蛋白+6 g/L琼脂+30 g/L蔗糖;愈伤组织最适分化培养基为MS+05 mg/L KT+01 mg/L NAA;最适生根培养基为1/2MS。
- Abstract:
- The experiment gets across using ten kinds of the hypocotyl and cotyledon of alfalfa as explants were used to study the effect of different mediums,hormones in different concentrations on callus induction rate and differentiation rate.The results indicated that the Medicago sativa L.cv.Guochan hypocotyl callus differentiation rate was the highest;the best medium for callus induction was the MS salt+UM vitamin,2.0 mg/L 2,4-D+025 mg/L KT,2.0 g/L casein acid hydrolysate+6 g/L Agar+30 g/L sucrose;the best differentiation medium was the MS medium+05 mg/L KT+01 mg/LNAA;the best root medium for plant development was 1/2 MS.
