新疆甘草根腐病病原鉴定及生物学特性测定

- 以2020年5—7月采集的甘草根腐病植株为试材,采用组织分离法分离病原菌并结合形态特征和分子生物学技术确定甘草根腐病的病原菌种类,研究了各种培养基、光、pH、温度、碳和氮源对病原菌生物学特性的影响,以期明确危害新疆甘草生长的根腐病病原菌种类和病原菌最适培养条件。结果表明:通过组织分离法分离出的96株真菌菌株,在科赫式法则中产生了与田间一致的发病症状,明确了新疆根腐病的病原菌是尖孢镰孢菌(Fusarium oxysporum)、腐皮镰刀菌(Fusarium solani)、轮枝镰刀菌(Fusarium verticillioides)。其最适培养条件为自然pH的PDA培养基接种,置于25~28 ℃光暗12 h交替恒温培养箱中培养。
- Abstract:
- Licorice root rot plants collected from May to July 2020 were used as test materials.Tissue separation technology was used to isolate pathogenic bacteria and combined with morphological characteristics and molecular biology techniques to determine the pathogen species of licorice root rot.Effects of various media,light,pH,temperature,carbon and nitrogen sources on biological characterisation of pathogenic bacteria were studied,in order to clarify the species of root rot pathogens and the optimum culture conditions of the pathogens that affect the growth of licorice in Xinjiang.The results showed that 96 fungal strains isolated by tissue separation method produced the same onset symptoms as those in the field according to Koch′s rule,and it was clear that the pathogens of root rot in Xinjiang were Fusarium oxysporum,Fusarium solani and Fusarium verticillioides.The optimal culture condition was PDA medium with natural pH and placed in a constant temperature incubator with alternating light and darkness for 12 hours at 25-28 ℃.
