- 以‘宁杞1号’‘宁杞菜1号’夏季的根、茎、叶、花鲜样为试材,采用高效液相色谱法及荧光定量PCR技术,研究了不同器官褪黑素含量及合成酶基因表达量,以期为后续宁夏枸杞褪黑素合成研究及药用价值挖掘提供参考依据。结果表明:‘宁杞菜1号’各器官褪黑素含量显著高于‘宁杞1号’,2个枸杞品种褪黑素含量均表现为花>叶>茎>根含量。TDC、T5H、COMT00、COMT01这4种酶基因在根、茎、叶、花中均有表达。
- Abstract:
- Taking the fresh roots,stems,leaves,and flowers of ‘Ningqi 1’ and ‘Ningqicai 1’ sampled in summer as the experimental materials,high-performance liquid chromatography (HPLC) and quantitative real-time polymerase chain reaction (qRT-PCR) were utilized to investigate melatonin contents and the expression profiles of melatonin biosynthetic genes across different organs,in order to provide reference for the subse quent research on the synthesis of melationin in Lycium barbarum L.and the exploration of its medicinal value.The results showed that the melatonin content in each organ of ‘Ningqicai 1’ was significantly higher than that of ‘Ningqi 1’.For both cultivars,the melatonin content followed the descending order of flower>leaf>stem>root.Additionally,the four biosynthetic genes,namely TDC,T5H,COMT00,and COMT01,were ubiquitously expressed in roots,stems,leaves,and flowers.
