青钱柳内生真菌分子鉴定及其抗菌研究

- 以广西百色野生青钱柳为试材,采用真菌形态学、rDNA-ITS、抗菌试验的方法,研究广西百色野生青钱柳内生真菌的遗传多样性和抗菌活性,以期了解广西百色青钱柳内生真菌的不同种群结构,筛选出具有抗菌活性的菌株。结果表明:从青钱柳植物中分离获得76株内生真菌,隶属于2门5纲12目19科25属,镰刀属(Fusarium)、拟盘多毛孢属(Pestalotiopsis)、间壳座属(Diaporthe) 为优势菌属;根、茎与韧皮部、木质部内生真菌多样性存在明显差异,茎部内生真菌的 Shannon index (H′=1.605 6)、根部(1.016 7),远高于韧皮部(0.209 7)和木质部(0.266 7)。5株内生真菌至少对 1 种病原指示菌有抑菌活性,其中,Pezicula sp.826对金黄色葡萄球菌和大肠杆菌的抑菌效果最强,Pezicula sp.927对热带假丝酵母菌的效果最强。综上可知,青钱柳植物中蕴藏着丰富多样的内生真菌资源及有较强抑菌活性的内生真菌,可为寻找新型抗菌物质提供宝贵资源。
- Abstract:
- The wild Cyclocarya paliurus (Batal.) Iljinsk in Baise,Guangxi was used as the test material,the genetic diversity and antibacterial activity of endophytic fungi from C.paliurus,in Baise Guangxi,were studied by means of fungal morphology,rDNA-ITS and antibacterial test,in order to understand the different population structure of endophytic fungi and screen out antibacterial activity of strains.The results showed that 76 strains,belonging to 25 genera,19 families,12 orders,5 classes,and 2 phyla were isolated from various tissues of C.paliurus,of which Fusarium、Pestalotiopsis and Diaporthe were the dominant genera.The diversity of endophytic fungi was significantly different among roots,stems,phloem and xylem,the Shannon index (H′=1.605 6) and root (1.016 7) of endophytic fungi in stem were much higher than in phloem (0.209 7) and xylem (0.266 7).The results of antibacterial activity showed that five strains of endophytic fungi inhibited at least one indicator pathogen.Among them,Pezicula sp.826 had the strongest inhibitory effect on Staphylococcus aureus and Escherichia coli,and Pezicula sp.927 had the strongest inhibitory effect on Candida tropicalis.According to the results of this study,there were abundant and diverse endophytic fungi and endophytic fungi with strong antibacterial activity in the precious medicinal plant C.paliurus,which could provide resources for the search of new antibacterial substances.
