- 以湖南大宗药材多花黄精种子为试材,研究不同消毒方法、单一激素对种子污染率和萌发率的影响,不同激素配比对种子芽生长和根系的诱导,以期建立多花黄精种子组培体系。结果表明:用8%NaClO消毒10~15 min配合0.1%HgCl2消毒10~15 min,污染率低,种子萌发率在55%~65%;温度为(26±2)℃,光照强度为2 000 lx条件下,每天光照12 h,当年采收的种子萌发率显著高于贮藏1年的种子,单一激素下种子的萌发率较低,且不同激素对种子萌发的诱导存在差异。激素配比能显著提高种子的萌发率,1.0 mg?L-1 6-BA+0.5 mg?L-1 NAA培养基组合有利于芽的生长;1.0 mg?L-1 IBA+0.5 mg?L-1 NAA培养基组合最有利于生根,培育周期8~10个月。该研究为多花黄精的育苗和品种改良提供一定技术支持。
- Abstract:
- The seeds of Polygonatum cyrtonema Hua were used as test materials to establish a high-efficient in vitro regeneration system.Effect of sterilization,single hormone on seed germination,and hormone ratio on bud growth and root induction were studied .The results showed that the optimal sterilization was using 8% NaClO for 10-15 minutes and 0.1% HgCl2 for 10-15 minutes was good forreducing pollution rate and germination rate between 55%-65%.The germination rate of seeds harvested in the same year was significantly higher than that of seeds stored for one year.Single hormone was not good for seeds gemination.The combination of 1.0 mg?L-1 6-BA+0.5 mg?L-1 NAA benefited to the elongation of adventitious buds.And 1.0 mg?L-1 IBA+0.5 mg?L-1 NAA was the best combination of hormones for rooting,and the temperature was (26±2)℃ with 2 000 lx light intensity for 12 hours one day.The cultivation period was 8-10 months.This study provided a technical support for seedling and variety improvement of removal of Polygonatum cyrtonema Hua.
