- 以NCBI核酸数据库中五味子DNA序列为研究对象,采用MEGA 6.0和DnaSP6软件分析法,研究了ITS2、PEPC、trnH-psbA和matK 4个目标片段的序列特征,以期获得五味子的遗传多样性和种质资源的来源。结果表明:ITS2序列的变异位点最多为15个;PEPC序列的单倍型最多为11个;最大遗传距离在ITS2序列之间为0.046;PEPC序列具有最高单倍型多样性为0.985,ITS2序列具有最高核苷酸多态性为0.004 93,所有序列的转换/颠换值均小于2;且4种序列构建的邻接法系统树结果均可以分为3个主要分支。
- Abstract:
- The DNA sequence of Schisandra chinensis Baill in the NCBI nucleic acid database were taken as the research object.The sequence characteristics of the four target fragments of ITS2,PEPC,trnH-psbA and matK were studied by using MEGA 6.0 and DnaSP6 software analysis methods,in order to obtain the genetic diversity and germplasm of Schisandra source of resources.The results showed that fifteen variable sites were found among ITS2 sequences;eleven haplotypes were found among PEPC sequences;the biggest genetic distance was 0.046 occur in ITS2 sequences;PEPC sequences owned highest haplotype diversity 0.985,ITS2 sequences owned highest nucleotide diversity 0.004 93,and all transition/transversion number of four fragments were less than two;and all phylogenetic trees based on four fragments were divided into three clades.
