枸杞新病害叶枯病症状表现及病原菌鉴定

- 为了明确枸杞新病害叶枯病的症状表现及病原菌种类,2015—2017年在甘肃省武威市凉州区长城乡具有代表性的枸杞种植园,对枸杞叶枯病的症状进行了系统调查,采用组织分离法和回接试验分离病原菌并测定其致病性,对病原菌显微形态及ITS序列进行了鉴定及分析。结果表明:枸杞叶和果实均可感染叶枯病菌,室内接种与田间回接和田间发病症状相同,叶子和果实的病原菌可以相互侵染,并在枸杞叶上和果上均表现出叶枯病典型症状;不同发病组织中分离纯化得到158株菌株,形态观察表明,侵染叶和果实的菌株均为链格孢属链格孢菌(Alternaria alternata (Fr.) Keissler)。通过ITS序列,并结合形态特征进一步确定引起枸杞不同组织叶枯病的病原菌为链格孢菌(A.alternate)。
- Abstract:
- In order to clarify the leaf blight disease symptom and pathogen species of Lycium baraum during 2015—2017,the leaf blight disease symptom was surveyed systematically in the representative L.bararum plantation where was located at Changcheng township,Wuwei,Gansu.The tissue separation and back-test were used to isolate the pathogen and determine its pathogenicity,and which was identified by morphological traits as well as sequence analysis of rDNA-ITS.The results showed that the samples from leaves and fruits of L.bararum were all able to be infected by the pathogen of leaf blight disease,and the symptom of indoor vaccination and field return was the same as in the filed.The pathogens of leaves and fruits could invade each other,and which showed the typical symptoms of leaf blight disease on leaves and fruits.The 158 strains were isolated from different samples,which were all identified as Alternaria alternata (Fr.) Keissler by morphological traits.By ITS sequence analysis and combined with morphological characteristics,it was further determined that the pathogen which caused leaf blight disease in different tissues of L.bararum was A.alternata.
