- 以当年生野生山杏成熟种子为试材,采用组织培养法,研究了种皮、不同浓度赤霉素(GA3)以及光照条件对山杏种子萌发的影响。结果表明:光照16 h、黑暗8 h条件下,MS培养基中添加 0.16 mg/L GA3〖KG-*3〗能有效促进去种皮山杏种子的萌发,未去除种皮的山杏种子均未萌发;在遮光条件下,去种皮种子在添加了0.16 mg/L与0.50 mg/L GA3〖KG-*3〗的MS培养基中发芽率均为100%,幼苗比光照16 h、黑暗8 h培养条件下的高且生长速度快,但黄化现象严重;将幼苗移至土壤中,能够正常生长,成活率达100%。
- Abstract:
- The mature seeds of prunus armeniaca was used as material to study the influence factors of embryo culture,including the seed testa,shading conditions and different concentration GA3.The results showed that the MS medium added 0.16 mg/L of GA3 could increase the germination of embryo removed testa under light period 16 h/8 h.Under the shading condition,the germination rate of embryo removed testa was 100% in MS medium added of 0.16 mg/L and 0.50 mg/L of GA3.The seedlings growth was faster than that of under light period 16 h/8 h,but the etiolation was obvious.The seedlings which were transplanted to soil were normal with rival rate 100%.
