- 以“红天鹅”蝴蝶兰为试材,研究了6苄氨基嘌噙(6-BA)浓度、取样节位、附加物对花梗腋芽诱导分化及不同外植体诱导原球茎分化的影响,以期建立蝴蝶兰组培快繁技术体系。结果表明:6-BA浓度为3.0 mg/L时,花梗腋芽分化率最高,为96%;取基部2、3节位幼嫩、粗壮的花梗腋芽诱导分化效果最好,分化率可达100%;以土豆替代香蕉、白砂糖替代蔗糖,可大大降低培养基的生产成本;根座和芽座是诱导原球茎分化的最佳外植体,诱导率可达90%以上。该试验筛选出了2条蝴蝶兰高效组培快繁技术体系。
- Abstract:
- Taking Doriaenopsis ‘Red Swan’ as material,in order to establish the tissue culture technique system of Phalaenopsis,the effect of 6-BA concentration,samples internodes,medium additives on peduncle axillary bud differentiation were studied,and the influence of different explants on peduncle axillary bud differentiation was also studied.The results indicated that the differentiation rate of peduncle axillary bud was up to 96% when the 6-BA concentration was 3.0 mg/L;the young but strong axillary bud,second or third internodes from the base of peduncle was the suitable explants to induction,the differentiation rate could reach 100%;replaced banana and sucrose by potato and white granulated sugar was an ideal method to reduce medium cost;generally levels of hormones were the highest in the base of root and bud stem,the rate of adventitious bud induction reached to 90%.Following that,two effective rapid propagation technique systems were found by series experiments.
