- 以苜蓿子叶节为外植体,以MS为基本培养基,研究了不同浓度激素对苜蓿离体再生体系中子叶节外植体分化、不定芽诱导、不定芽伸长及生根的影响,以期建立高效的苜蓿子叶节离体再生体系。结果表明:苜蓿子叶节再生最佳萌发培养基为MS;最佳诱导培养基为MS+1.0 mg/L 6BA,诱导时间为7~10d;不定芽伸长的最佳培养基为MS+0.5 mg/L 6BA;最佳生根培养基为1/2MS+0.1 mg/L IBA,15 d即可生根。
- Abstract:
- Taking the cotyledonary nodes of Medicago sativa as explants and MS as the basic medium,the effect of different proportion of hormones on explants differentiation,adventitious bud induction,elongation and rooting were studied.The results showed that the optimum medium for alfalfa cotyledon node regeneration:germination medium was MS;the best induction medium was MS+1.0 mg/L 6BA,with induction time 7 to 10 d;the best adventitious shoot elongation medium was MS+0.5 mg/L 6BA;the best rooting medium was 1/2MS+0.1 mg/L IBA,root could be formed in 15 days.
