- 为了明确分布在山西省不同地域的野罂粟材料之间的亲缘关系和遗传多样性,以在山西省采集的6份野罂粟和采集于内蒙古的1份野罂粟引种为研究对象,利用改进的CTAB法提取DNA,并通过单因素多水平梯度试验,筛选了模板、Mg2+、Taq DNA聚合酶、dNTP和随机引物的浓度及用量,建立了山西省野罂粟RAPD技术扩增体系。结果表明:在20 μL反应体系中,Taq DNA聚合酶、模板、引物、Mg2+和dNTP 5种主要成分的适宜浓度和用量分别为:2.0 U、20 ng、1.2 μmol/L,2.0 mmol/L、150 μmol/L,并在此基础上构建UMPGA聚类图,经过分析得知,地域分布为野罂粟聚类的主导因素,花色为野罂粟聚类的次要因素。
- Abstract:
- To provide bases of establishing Papaver nudicarule genetic relationship and genetic diverslty,taking 7 Papaver nudicarule collected and preserved from Shanxi and Inner Mongolia as test materials, DNA was extracted by improved CTAB and concentration and dosage of template,dNTP,Mg2+,Taq DNA polymerase,dNTP and primer were screened by single-factor multi-level gradient experiment.The experiment established RAPD technology amplification system on Papaver nudicarule in Shanxi Province.The results showed that the optimized RAPD system was Taq DNA polymerase 2.0 U,template DNA 20 ng,primer 1.2 μmol/L,Mg2+ 2.0 mmol/L and dNTP 150 μmol/L in total volume 20 μL amplification system and on this basis builded UMPGA dendrogram.Clustering results showed that geographical distribution of Papaver nudicarule was the leading factor,and the flower color was a secondary element.
