- 以羊肚菌深层发酵后除去菌丝体的滤液为原料,在单因素试验的基础上,采用正交实验设计,研究了乙醇浸提羊肚菌胞外多糖和超声波辅助提取菌丝体多糖的最佳方法。结果表明:乙醇浸提羊肚菌胞外多糖最佳条件为沉淀温度-15 ℃,加乙醇3倍量,沉淀时间24 h,沉淀pH 7;超声波辅助提取菌丝体多糖最佳条件为超声波破壁时间15 min,超声波功率400 W,料液比1∶10 g·mL-1,提取次数2次。
- Abstract:
- After the deep fermentation of Morchella mycelia,the filtrate removed mycelia was used as raw material.On the basis of single-factor experiments,the orthogonal test was used to study the best method for the extraction of extracellular polysaccharide from Morchella by ethanol.The results showed that the optimum conditions for extracting extracellular polysaccharides from Morchella by ethanol was precipitation temperature -15 ℃,ethanol 3 times,precipitation time 24 hours,precipitation pH 7.The optimal conditions for ultrasonic-assisted extraction of mycelium polysaccharides were ultrasonic breaking time of 15 minutes,ultrasonic power of 400 W,ratio of material to liquid of 1∶10 g·mL-1,and extraction times of 2 times.
