滇龙胆种质资源筛选及离体培养的初步研究

- 以不同居群、同一居群不同滇龙胆植株为试材,采用HPLC法对滇龙胆根茎中龙胆苦苷含量进行测定,再以龙胆苦苷含量≥8.0%的感通、湾桥和蝴蝶泉居群茎尖为外植体,研究了培养基、培养温度对滇龙胆离体培养及植株再生的影响。结果表明:大理苍山滇龙胆不同居群根茎中龙胆苦苷含量差异达到显著或极显著水平,其中,感通居群、中和居群、湾桥居群和蝴蝶泉居群根茎中龙胆苦苷含量较高,达到8.0%以上;上关居群根茎中龙胆苦苷含量最低,为(5.849±0.01493)%。滇龙胆同一居群不同植株根茎中龙胆苦苷含量差异较小,喜洲居群不同植株根茎中龙胆苦苷含量差异达到显著水平(P<0.05),中和居群不同植株根茎中龙胆苦苷含量差异达到极显著水平(P<0.01),其它居群不同植株根茎中龙胆苦苷含量无显著差异。滇龙胆茎尖生长点离体培养及植株再生的最适宜培养基为1/2MS培养基,再生植株诱导率可达80.00%,不定芽高(5.6±0.2)cm,最佳培养条件是20℃/25℃变温培养,再生植株诱导率可达87.78%,不定芽高(7.5±0.2)cm。
- Abstract:
- Taking different population and the different individuals within the same population of Gentiana rigescens Franch as materials,the gentiopicroside content in the rhizome of Gentiana rigescens Franch was determined by HPLC method,then the high quality germplasm resources of which the gentiopicroside content was greater than or equal to 8.0% were selected as the explants,and the effects of culture medium and culture temperature on the in vitro propagation and plant regeneration of Gentiana rigescens Franch were explored.The results showed that the gentiopicroside content in rhizome of Gentiana rigescens Franch from different populations of Dali Cangshan reached significant or extremely significant level.Among them,in Gantong population,Zhonghe population,Wanqiao population,and Hudiequan population,the gentiopicroside contents were high,and the content reached more than 8.0%.In Shangguan population,the gentiopicroside content was the lowest (5.849±0.0149%).Within the same population,the gentiopicroside contents of individual plant in rhizome of G.rigescens had small difference.The difference in Xizhou population reached significant level (P<0.05),in Zhonghe population reached extremely significant level (P<0.01),while in other populations,the gentiopicroside contents had no significant difference.The most suitable culture medium for stem-tip growth-point in vitro propagation and plant regeneration of Gentiana rigescens Franch was 1/2MS,and the induction frequency of plant regeneration could reach 80.00%,adventitious buds height (5.6±0.2) cm.The optimal culture condition was variable temperature culture at 20/25℃,and the regenerated plant induction frequency could reach 87.78%,with adventitious buds height (7.5±0.2) cm.
