- 以一品红带芽茎段、叶片为外植体,研究了不同激素浓度配比的MS培养基对愈伤组织、芽的分化及增殖和试管苗生根的不同效应。结果表明: 最佳愈伤组织诱导培养基为MS+BA 02 mg/L+NAA 05 mg/L+蔗糖30 g/L+琼脂75 g/L (pH 56),诱导率达到925%;芽的分化最佳培养基为MS+BA 20 mg/L+NAA 02 mg/L+蔗糖30 g/L+琼脂75 g/L,不定芽的诱导率达到833%;芽的增殖培养基为MS+BA 10 mg/L+NAA 02 mg/L+蔗糖30 g/L+琼脂5 g/L,增殖系数为108;而最佳生根培养基为1/2 MS+BA 02 mg/L+NAA 05 mg/L+蔗糖30 g/L+琼脂75 g/L,每芽生根数平均为35条。该研究结果为一品红组培苗的工厂化生产提供了可靠的技术指标。
- Abstract:
- Using leaf and stem with axilla bud of Euphorbia pulcherrima Wild as the explant respectively, effects on the callus induction,the shoot differentiation and propagation,and the root differentiation of different phytohormone combition in MS basic medium were studied.The results showed that the best medium for callus induction was MS+BA 02 mg/L+NAA 05 mg/L,and the differentiation rate reached 925 %;The medium MS+BA 20 mg/L+NAA 02 mg/Lwas the most suitable for shoot induction than others,and the differentiation rate reached 833%;The medium with MS+BA 10 mg/L+NAA 02 mg/Lwas the best one for shoot propagation,the propagation rate was 108;Whereas,the best medium for root differentiation was 1/2 MS+BA 02 mg/L+NAA 05 mg/L,and the average root number per shoot was 35.This result would was beneficial to culture the Euphorbia pulcherrima Wild seedlings industrially.
