- 以桑黄为试材,通过对比试验,对桑黄干菇组织分离的过程进行优化,以期提高分离成功率。结果表明:提前用水对组织块浸泡2~3 h,用75%酒精浸泡杀菌15~30 s,PDA培养基中加入抗生素,可以大幅度提高分离成功率,达到80%以上,甚至高达100%。
- Abstract:
- The tissue separation process of dried mushroom of Phellinus igniarius was optimized through comparative experiments with Phellinus igniarius as the experimental material,in order to improve the separation success rate.The results showed that through the comparison of this experiment,the process of tissue isolation of dried mushroom Phellinus igniarius was optimized.It could be seen that the tissue block was soaked in water for 2-3 hours in advance,the tissue block was soaked in 75% alcohol for sterilization for 15-30 seconds,and antibiotics were added in PDA medium,which could greatly improve the success rate of separation,which could reach 80% or even up to 100%.
