- 以侧柏优良无性系‘立叶’侧柏和良种‘蝶叶’侧柏一年生半木质化枝条为试材,采用完全随机处理研究速蘸(10 s)和浸泡(4 h)处理下不同植物生长调节剂(IBA、NAA、m (IBA)∶m (NAA)=1∶1、根旺)、不同浓度(100、200、300、400、600、800、1 000、1 500、2 000、2 500、3 500、4 500 mg·L-1)对侧柏扦插繁殖生根效果的影响,并运用隶属函数法对各处理的育苗效果进行综合评价,以筛选出侧柏扦插人工诱导生根的最佳处理组合,提高育苗效果及效率,同时采用石蜡切片技术观察生根过程中插穗基部组织结构变化,从解剖学角度揭示侧柏扦插生根过程中插穗内部结构变化和不定根的形成。结果表明:‘立叶’侧柏、‘蝶叶’侧柏经2 000 mg·L-1 IBA速蘸10 s处理后生根率最高,分别为92.22%和56.67%;隶属函数综合评价表明,800 mg·L-1 IBA浸泡4 h和2 000 mg·L-1 IBA速蘸10 s分别为‘立叶’侧柏、‘蝶叶’侧柏扦插育苗的最佳组合。解剖学结果显示,侧柏根原基属诱导根原基,不定根多发生于愈伤组织,少数不定根发生于髓射线与韧皮部交叉处,扦插生根可持续至100 d左右。
- Abstract:
- With one-year-old semi-lignified cutting of P.orientalis ‘Liye’ and P.orientalis ‘Dieye’ as materials,the effects of different plant growth regulators (including IBA,NAA,m (IBA)∶m (NAA)=1∶1 and GW) and its different concentration levels (including 100,200,300,400,600,800,1 000,1 500,2 000,2 500,3 500,4 500 mg·L-1) on rooting of cuttings by completely random combination test under rapid dipping (10 seconds) and soaking (4 hours) treatments were studied,and seedling effects of different treatments were comprehensively evaluated by membership function method.The aim was to choose the best combination of rooting treatments to improve the seedling raising effect and efficiency.At the same time,the tissue structure of cutting base was observed during the rooting process using paraffin sectioning technology,and the internal structure changes and adventitious root formation of cuttings during the rooting process were revealed.The results showed that the highest rooting rate of P.orientalis ‘Liye’ and P.orientalis ‘Dieye’ were 92.22% and 56.67% after treatment with 2 000 mg·L-1 IBA for 10 seconds.The comprehensive evaluation of membership functions showed that 800 mg·L-1 IBA immersion for 4 hours was the best combination for cutting seedlings of Platycladus orientalis ‘Liye’,and 2 000 mg·L-1 IBA quick dipping for 10 seconds was the best combination of P.orientalis ‘Dieye’ cuttings.Anatomical results showed that the root primordia of P.orientalis was produced by induction,and adventitious roots occurred mostly in callus.A few adventitious roots occurred at the intersection of pith ray and phloem,and the rooting cycle last about 100 days.
