- 以采集于秦巴山区的野生麦冬根、叶及花为试材,采用单因素和响应面试验的方法,研究了超声辅助提取麦冬花多酚的提取工艺并评价了麦冬不同部位的多酚含量及其抗氧化功效,以期优化麦冬不同组织类多酚提取的工艺参数。结果表明:麦冬花多酚的最佳提取工艺条件为超声时间31 min,提取温度57 ℃,乙醇浓度63%,料液比1∶19 g·mL-1,其多酚提取量(32.95±0.14)mg·g-1,麦冬块根、叶及须根的多酚含量分别为(24.64±0.05)、(20.52±0.04)、(17.42±0.03)mg·g-1。与常规抗氧化剂维生素C对比,不同部位的麦冬多酚均具有较好地清除O·[TX-*4]2、·OH、NaNO2的能力,其总体清除活性顺序为麦冬花>麦冬块根>麦冬叶>麦冬须根≈抗坏血酸;麦冬花多酚对O·[TX-*4]2、·OH和NaNO2的清除IC50值分别为(0.41±0.01)、(0.097±0.005)、(0.021±0.005)mg·mL-1,而同浓度条件下维生素C溶液的清除IC50值分别为(0.80±0.01)、(0.14±0.01)、(0.012±0.002)mg·mL-1,表明不同部位的麦冬多酚均具有相对较强的抗氧化活性。
- Abstract:
- With single factor and response surface method,this study investigated the optimal process parameters for ultrasonic-assisted extracting total polyphenols from Ophiopogon japonicus flowers,which was sampled from the Qinba Mountains.Meanwhile,the scavenging effects of polyphenols from different parts of Ophiopogon japonicus on superoxide anion radical(O·[TX-*4]2),hydroxyl free radical(·OH) and nitrite were also examined.The results showed that the optimum condition for ultrasonic-assisted extracting of polyphenols from Ophiopogon japonicus flowers were as follows,extraction temperature 57 ℃,ethanol concentration 63%,ultrasonic time 31 minutes,ratio of material to liquid 1∶19 g·mL-1).Under this condition,the content of polyphenols from Ophiopogon japonicus flowers was (32.95±0.14)mg·g-1.The polyphenols extraction contents from Ophiopogon japonicus tuber roots,leaf and fibrous root were (24.64±0.05)mg·g-1,(20.52±0.04)mg·g-1 and (17.42±0.03)mg·g-1,respectively.Comparing with conventional antioxidant ascorbic acid,total polyphenols from different parts of Ophiopogon japonicas extraction could be better removing superoxide anion radical(O·[TX-*4]2),hydroxyl free radical(·OH) and nitrite,and the order of scavenging free radicals was as follows:Ophiopogon japonicus flowers>tuberous root>leaf>fibrous root≈ascorbic acid.The polyphenols from Ophiopogon japonicus flowers had certain scavenging ability on O·[TX-*4]2,·OH and NaNO2 with IC50 of (0.41±0.01)mg·mL-1,(0.097±0.005)mg·mL-1and (0.021±0.005)mg·mL-1,respectively,and scavenging free radical IC50 of vitamin C at the same concentration for O·[TX-*4]2,·OH and NaNO2 were (0.80±0.01)mg·mL-1,(0.14±0.01)mg·mL-1 and (0.012±0.002)mg·mL-1,respectively.The study proved that polyphenols from different parts of Ophiopogon japonicus extraction had relatively good antioxidant activity.
