- 以‘春绿兰×虎雪兰’F1代的原球茎为外植体,在无菌条件下,以1/2MS培养基为基本培养基,添加不同浓度6-BA与NAA,研究了不同激素配比对原球茎不同培养阶段的增殖、分化和生根的影响。结果表明:1/2MS+1.0 mg?L-1 6-BA+0.5 mg?L-1 NAA+80 g?L-1香蕉泥,利于原球茎的增殖和分化,增殖率和分化率分别为222.00%、90.57%;1/2MS+1.5 mg?L-1 6-BA+0.5 mg?L-1 NAA+80 g?L-1香蕉泥,利于不定芽增殖,增殖率为42.50%;1/2MS+0.5 mg?L-1 6-BA+1.5 mg?L-1 NAA生根效果最佳,生根率为100%,平均株根数为5.50条,平均根长为40.73 mm,且植株长势健壮。
- Abstract:
- The Cymbidium faberi Rolfe var.Szechuanicum×(C.tracyanum var.Huanghua×C.mastersii) F1 hybrids as test material,protocorm-like-body (PLB) were used as explants in tissue culture to study the influence of BA and NAA in different concentrations,mashed bananas and activated carbon on their regeneration under 1/2MS basic medium.The results showed that 1/2MS+1.0 mg?L-1 6-BA+0.5 mg?L-1 NAA+80 g?L-1mashed bananas could make PLB rate to 222.00% and differentiation rate to 90.57%,1/2MS+1.5 mg?L-1 6-BA+0.5 mg?L-1 NAA+80 g?L-1 mashed bananas could make multiplication rate to 42.50%,1/2MS+0.5 mg?L-1 6-BA+1.5 mg?L-1 NAA could make rooting rate to 100%,the average number of plant roots was 5.50,average root length was 40.73 mm,and plant grew healthily.
