- 以2个黄瓜品种“金谷六号”和“吉列168”的种子为试材,以子叶和下胚轴作为外植体进行离体培养,研究了不同培养基和激素浓度及配比对黄瓜愈伤组织诱导、不定芽分化的影响。结果表明:2种不同基因型的黄瓜均表现出较强的愈伤组织分化能力,下胚轴要优于子叶,不同激素配比组合对愈伤组织的形态影响较大,AgNO3能有效抑制愈伤组织的玻璃化。2种黄瓜以子叶为外植体的最佳激素配比均为MS+0.5 mg?L-1 6-BA+0.10 mg?L-1 IAA+1.0 mg?L-1 AgNO3;2种黄瓜以下胚轴为外植体的最佳激素配比均为MS+2.0 mg?L-1 6-BA+0.10 mg?L-1 IAA+1.0 mg?L-1 AgNO3。该试验条件下不定芽的分化没有形成。
- Abstract:
- Two different kinds of cucumbers,‘Jingu six’ and ‘Gillette 168’ were selected as the materials,cotyledon and hypocotyl as explants,the effect of culture medium type,hormone concentration,hormone ratio on callus induction and differentiation of adventitious bud were investigated.The results showed that both cucumbers showed the strong callus differentiation ability,and hypocotyl was superior to cotyledon;the shape of the callus was quite different under different hormone ratios.AgNO3 as the additives could commendably restrain the vitrification of the callus induction;both cucumbers had the same best hormone ratio for cotyledon explant MS+0.5 mg?L-1 6-BA+0.10 mg?L-1 IAA+1.0 mg?L-1 AgNO3 and for hypocotyl explant MS+2.0 mg?L-1 6-BA+0.10 mg?L-1 IAA+1.0 mg?L-1 AgNO3.The differentiation of adventitious bud was not formed.
