- 以宁夏枸杞新优品系为试材,利用花药培养技术,研究了不同基因型、激素配比、低温预处理、热激处理、添加活性炭对其胚状体诱导率的影响。结果表明:宁夏枸杞新优品系‘06-16’在基因型筛选中,诱导率最高为0.95%,通过对试验体系进行优化,2.0 mg/L KT与0.2 mg/L 2,4-D 的组合诱导率达到2.07%。4℃低温预处理3 d或5 d、33℃热激处理5 d、活性炭浓度为0.8%,均能提高新优品系‘09-106’的胚状体诱导率。
- Abstract:
- Taking Ningxia wolfberry new superior strain as the materials,using anther culture technology,the effect of different genotypes,hormone ratio,cold and hot temperature pretreatment,carbon on embryoid induction from the anther of Ningxia wolfberry was studied.The results showed that the ‘06-16’ was easy to be induced.The medium adding 2.0 mg/L KT and 0.2 mg/L 2,4-D was suitable for the induction of embryoid,the induction rate was 2.07%.When the flower bud was treated 3 days or 5 days under 4℃,cultivated in the high temperature (33℃),adding 0.8% active carbon in midia,the induced rate could be improved.
