- 以“晶瑶”草莓叶片为外植体,研究了不同激素配比、AgNO3浓度和暗培养对草莓叶片不定芽分化的影响。结果表明:当培养基为 MS+噻二唑苯基脲(TDZ) 1.0 mg/L+吲哚丁酸(IBA) 0.4 mg/L+AgNO3 2.0 mg/L时,不定芽再生率高达70.3%,每个外植体平均再生芽数为4.2个;AgNO3可以有效抑制“晶瑶”草莓叶片玻璃化和褐变;暗培养处理促进“晶瑶”草莓叶片的不定芽诱导,最佳暗培养时间为14 d。
- Abstract:
- Taking the leaves of strawberry ‘Jingyao’ as explants,the effect of different hormone combination,AgNO3 and dark culture on adventitious bud were investigated.The results showed that the optimum medium for adventitious bud regeneration was MS supplenmented with 1.0 mg/L thidiazuron(TDZ),0.4 mg/L indole-3-butyric acid(IBA) and 2.0 mg/LAgNO3,under this conditions,the isolated regeneration rate of adventitious bud from leaf explant reached 70.3%,and the average number of regenerated buds reached 4.2;AgNO3 could effectively restrain strawberry leaf vitrification and brown stain;dark treatment promoted adventitious bud differentiation of strawberry leafs,the best time of dark treatment was 14 d.
